双重组酶介导的 cassette 交换(dRMCE)被用来将内源基因的 ex1 编码区域替换为带有 flag 标签的 ex1。未经修改的 ex2 替换了内源的 ex2，同时插入了 attB/attP 包裹的 PGK-hygro 选择性载体。在 loxP 突变体的 Hand2 诱导性多能干细胞中进行了这种改造，ex1 和 ex2 之间有 FRT 包裹的 neomycin 遗传元件位于下游。替换过程通过 ES 细胞中同时表达 cre 和 flp 重组酶来实现。(来源：J:201720)
