设计了一个靶向载体，它包含一个内源性核糖体进入位点(IRES)连接到CreERTM融合基因(CreERTM，是Cre重组酶与小鼠雌激素受体配体结合区域的G525R突变形式)，并将这个CreERTM序列插入Myf5基因第3外显子的终止密码子之后。载体中插入了由Frt座点 flank的诺卡霉素抗性标记。这个构建通过电穿孔法导入R1胚胎干细胞。经过定向的ES细胞被注入C57BL/6囊胚，然后杂合雄性与C57BL/6雌性进行交配。(来源：J:199482)
