在2号外显子下游插入了一个FRT引导的neo cassette，同时选择 cassette 也通过loxP座进行 flank。通过使用Flp重组酶，从靶向恰当的鼠中剔除了选择 cassette。随后，通过Cre表达去除2号外显子，通过RT-PCR检测到了剪接的mRNA片段，但 Western blot 检测并未发现蛋白质。(来源：199861)
