使用重组融合技术构建了一个靶向载体，包含Cre重组酶连接SV40poly(A)信号，下游是Frt-介导的Pgk-neo片段。随后，对含有Clec9a基因的BAC（RP23-248K14）进行了改造，通过Flp重组酶去除了neo标记。改造后的BAC经过纯化，用于核移植以产生转基因动物。目前找不到供体动物的祖先信息。（来源：J:199355）
