设计了一种靶向载体，目标是将cre/ERT2-SV40 polyA插件序列，后跟一个FRT5引导的pSV40-NeoR-pA片段，插入活动调节的细胞骨架相关蛋白基因(Arc)的起始翻译位点。这个操作移除了内含子和编码区域，并用外源SV40的polyA信号替换了内源性3' UTR（这影响mRNA的稳定性并参与Arc mRNA在树突中的运输）。所有起始翻译位点前的序列都保留了下来。生成的转基因小鼠被繁殖到能激活GFP-FlpO转基因小鼠的生殖细胞中，以删除FRT5引导的pSV40-NeoR-pA片段。(来源：J:196644)
