使用一个包含IRES-PLAP-IRES-lacZ（IRES是内源性核糖体进入位点，PLAP是胎盘碱性磷酸酶，lacZ有核定位信号）序列和一个自我剪切的neo抗性元件（ACN）的靶向构建，来针对小鼠Pgr基因在ES细胞中。将ES细胞注入囊胚以产生嵌合体。ACN cassette中的neo基因在通过雄性生殖系统时自我剪切，因此杂交雄性后代与野生型C57BL/6J雌性杂交产生的F1后代会有ACN的缺失。（来源：J:196278）
