设计了一个构建，包含内源性核糖体进入位点（IRES）、Cre重组酶基因、Frt位点、RNA聚合酶II-Neo-HSVtk-polyA插接片段，以及孕酮受体（Pgr）转录终止密码子下游的第二个Frt位点。这个构建通过电穿孔法导入了129S/SvEv衍生的胚胎干细胞（ES细胞）。经过定向的ES细胞被注入受体囊胚中。杂交雄性与C57BL/6J雌性进行交配，以建立群体。随后，突变小鼠与GtROSA-FLPe（未透露遗传背景）小鼠杂交，以剔除含Frt-flanked Neo cassette的片段。（来源：J:196278）
