设计的靶向载体包含从5'至3'的合成剪接接受器、一个loxP位点、一个起始密码子（ATG）、野生型H3.3型（H3f3a）基因编码序列、H3家族3A的3' UTR以及SV40的polyA序列。随后是下游带有frt标记的诺卡因抗性基因（neo cassette）和第二个loxP位点，接着是起始密码子、黄色荧光蛋白（YFP）的venus变异体，以及另一个SV40的polyA序列。这个载体替换了解码子2的一部分。通过Flp介导的重组移除了neo cassette，而Cre介导的重组则去除了H3.3异构体，使得报告基因得以表达。（来源：J:194069）
