在BAC中，通过同源重组技术，将一个位于Nfatc1外显子1起始翻译位点的核定位反四环素转录激活序列和Frt内含子标记的PGK-诺卡因 cassette插入。随后，通过Flpe酶切去除neo cassette。纯化后的构建被注入FVB受精卵，第0.5日的囊胚阶段。获得了7个杂交瘤系，这些系与tetO-cre;Gt(ROSA)26Sorfslz小鼠杂交，随后在E7.5开始，通过3天的多西环素诱导。三个系（96、100和122）在心内膜及其衍生的心内膜垫中显示了表达。具体信息见原始参考文献(#96)。
