在翻译起始密码子下游，插入了一个带有3' loxP位点的FRT引导的neo cassette。接着在终止密码子下游，再插入了一个loxP位点。通过Flp介导的重组，neo cassette被去除，大部分编码序列被留下了可剪接的区域。随后，Cre介导的重组则移除了大部分编码序列。（来源：J:193058）
