在野生型exon 20上游有一个loxP位点，后面跟着一个polyadenylation序列，接着是一个FRT标记的neo cassette，4个转录终止子，再是另一个loxP位点，以及一个修改过的exon 20。修改过的exon 20在第1047位由A替换为G，导致了赖氨酸（H1047R）替换为histidine。通过Flp介导的重组移除了FRT标记的neo cassette。要删除野生型exon 20并表达修改过的exon 20，需要Cre介导的重组。(来源：J:193377)
