在编码外显子的第30个核苷酸位置，插入了COIN模块(包含L71 loxP位点、倒置poly(A)序列、倒置的EGFP、L66 loxP位点和FRT引导的neo cassette)。这个插入导致了CpG岛的分割。通过Flp介导的重组，neo cassette被去除了。要表达EGFP，需要Cre介导的重组。逆转录PCR确认Sox2ot和Mir1897的正常转录表达。(来源：J:191980)
