这个构建包含了一个5.8千碱基的nestin启动子片段，带有优化的phiC31重组酶( phiC31*，构建中的编码序列去除了核定位信号)，下游是tamoxifen诱导的突变雌激素受体(ERT2)序列，接着是1.8千碱基的nestin内含子，包含神经干细胞/前体细胞特异的增强子。SV40的polyadenylation序列被克隆到了载体中。经过纯化的构建被注入了受精的单细胞胚胎中。建立了7个克隆，其中2个(线号2和4)因在发育神经系统中ventricular zone的高转录基因表达，被选中进行进一步研究。(来源：J:185547)
