通过ES细胞的同源重组，靶向载体将一个含诺卡因（neo）抗性基因座插入Relb基因的第4内含子，并在第4外显子上游184bp位置插入一个loxP位点。随后，通过在ES细胞中转染表达Cre重组酶的质粒(pMC-cre)，短暂地去除了neo基因座，从而产生了第4外显子被敲除的等位基因。参考文献：J:169863, J:188293, J:270571
