使用了Pdx1tm1Mgn的重组酶介导 cassette 交换（RMCE）技术，插入了一个FRT标记的hygro cassette在区域IV的上游，同时将核定位的CFP插入到起始密码子位置，替换部分exon。通过Flp介导的重组，移除了hygro cassette。区域IV的上游保留了一个lox66/71位点，而CFP的下游则有一个lox2272位点。（来源：J:187872）
