Cre重组酶被插入到Etl4（Skt）基因座的第14个内含子中，通过使用带有左元件/右元件(LE/RE)突变Lox站点的可交换基因陷阱系统，实现了通过重组酶介导的 cassette exchange。原本陷阱载体中的IRES-cre序列被替换成了IRES-lacZ-neo功能单元，通过将载体和pCAGGS-cre质粒一起电穿孔到含被陷阱的Etl4基因的TT2 ES细胞中实现这一操作。(来源：J:187217)
