设计了一种靶向载体，其目的是在exon 1的起始密码子上游插入一个loxP位点。载体中包含了一个frt标签的诺卡因抗性基因(neo)区和第二个loxP位点，这些都位于exon 1之后。杂合子小鼠与Flp转基因小鼠杂交，以删除neo cassette(来源：J:187145)。
