将icre序列插入到了一个BAC（RP23-343C17）中，通过重组修复技术，替换了gcg（胰高血糖素；胰岛素前体）基因座2号外显子起始密码子和6号外显子终止密码子之间的序列。修改后的BAC经过纯化后，被注入了成熟的卵细胞。最初建立了四个祖先系，拷贝数进行了测定（线1：01，7-8个拷贝；12：2-3个；14：1个；30：1个）。详细研究了线12（J:186436）。
