通过将0.9kb的Col2a1启动子片段、4个串联的成骨特异性Acan增强子、一个内含子的切接接受体、反式四环素激活器编码序列和一个polyadenylation位点组成，构建了一个双基因构造。一个4kb的Trp53内含子绝缘序列将rtTA序列与构造的其余部分隔开，这部分包括7个串联的四环素响应元件(tetO)、人巨细胞病毒启动子(hCMV)、Cre重组酶编码序列以及一个polyadenylation位点。目前，转基因小鼠的创始信息尚不可用。在给予多西环素治疗后，rtTA被激活并结合tetO，从而诱导Cre表达。(来源：J:183084)
