将绿色荧光蛋白（EGFP）的编码序列后面跟着一个polyadenylationsignal插入了细菌人工染色体RP23-102G4中Zfp423的起始密码子位置，位于exon 1。修改后的BAC不含Zfp423的任何功能区域，也不包含任何其他注释基因的全长编码序列。产生了五个克隆系，其中两个在表达EGFP的细胞中Zfp423 mRNA丰度显著增加。其中一条克隆的数据已发表（文献来源：J:182285）。
