C57BL/6小鼠的BAC（Bacterial Artificial Chromosome）RP24-88H21经过改造，插入了只包含Slc17a8基因座的区域。改造过程中，使用了优化的cre重组酶（icre）和SV40的polyA信号，插入到了Slc17a8基因的起始ATG位置。这个修改后的BAC被注入到FVB/N雌性卵母细胞中，成功产生了转基因个体，随后与表达cre依赖性报告基因的实验鼠杂交。cre的活性在海马、皮质、纹状体和小脑的神经元中可见，同时也出现在视网膜内层的视锥细胞中。四个创始系中有两个表达cre，其中一条线路被进一步研究。（来源：J:182798）
