在第1外显子的起始翻译位点上游，插入了一个带有5' loxP位点的FRT引导的neo cassette。在第2外显子下游，又插入了一个loxP位点。通过Cre介导的重组，neo cassette、第1外显子起始密码子下游的3'端、内含子1以及第2外显子都被去除了。 Western blot结果显示胸腺细胞中没有蛋白质表达。（来源：J:177782）
