原初的OT-I转基因构建来自F. Carbone（墨尔本大学，维多利亚，澳大利亚）。OT-Iα链通过C端连接一个20个氨基酸的柔性连接器和增强型GFP，使用标准分子克隆技术表达，由CD4增强子和启动子驱动，但不包含沉默子。OT-Iβ链则在其内源性启动子的驱动下表达。（J:177978）
