RMCE通过重组酶介导的 cassette exchange 技术，将 Gt(ROSA)26Sortm1Maoh 基因中的 JT15 和 lox2272 之间的表达 cassette 替换为：从 5' 至 3'，包含 IRES、lacZ、polyA序列、CAG启动子、Hygro基因、polyA序列、FRT序列、polyA序列，以及由CAG驱动的EGFP和针对Tyrosinase的短发卡宾RNA融合。Tyrosinase基因内的RNAi靶序列是 "ctggaaggatttgccagtccac" 和 "gaactgccaatttcagcttta"。Flp介导的重组会去除除了FRT序列和lox2272位点周边的CAG启动子RNAi构建。来源：(J:173808)
