一个包含FRT位点介导的NEO插件和IRES-MerCreMer序列的靶向载体被插入到Oct4/Pou5f1基因的3' UTR区域。这个构建在InGenious Targeting Laboratory的C57BL/6来源的胚胎干细胞中通过电穿孔技术导入。经过正确筛选的ES细胞被注入到B6(Cg)-Tyrc-2J/J或BALB/c的囊胚中。产生的嵌合动物随后与B6.Cg-Tg(ACTFLPe)9205Dym/J小鼠杂交，以去除FRT位点周围的筛选 cassette。(参考文献: J:194647)
