设计了一个靶向载体，用于将CreERT2融合基因（Cre-ERT2，人雌激素受体配体结合域的G400V/M543A/L544A三重突变）、SV40polyA信号和FRT引导的PGK-Neo cassette插入Cux2基因的起始密码子位置。这个载体通过电穿孔技术导入了C57BL/6背景的Bruce4胚胎干细胞。杂交小鼠与C57BL/6纯系鼠杂交以建立群体。随后，突变小鼠与表达FLPe的C57BL/6纯系背景鼠杂交，以剔除FRT引导的PGK-Neo cassette。最终，Cux2-CreERT2小鼠与C57BL/6J纯合小鼠杂交三代左右（以去除FLPe转基因），(J:151755)。
