将含有一整套Phox2b基因座以及Phox2b基因座上游130kb序列（包括Limch1基因区段）和下游70kb序列的鼠标BAC RP24-95M11改造，构建了表达Cre重组酶的构造。通过同源重组/BAC重组技术，将带有Frt-flanked Neo标记的Cre重组酶序列插入到Phox2b基因座的起始ATG位置。在BAC重组过程中，通过转染表达FLP的载体，Frt-flanked Neo标记被去除。最终得到约200kb的Phox2b-Cre BAC转基因，用于C57BL/6J小鼠的核移植，产生了独立的祖先系。在这些祖先系中，祖先系3的孤束核（NTS细胞）中Cre重组酶表达量最高，相关信息参考文献为(J:172264)。
