使用一个包含Sox10基因座完整序列的120kb基因组PAC片段构建了转基因。Sox10基因的3-5号外显子区域通过同源重组在细菌中用loxP引导的eGFPpolyA插件替换，后面跟着编码减毒G383A型白喉毒素亚单位A(DTA)的序列。为了筛选，包含Frt标记的Cm cassette被包含进来，通过暂时激活Flp在细菌细胞中去除。在Cre重组酶存在的情况下，报告基因片段会被剔除，从而驱动DTA表达并导致表达细胞的消除。(来源：J:105334)
