设计了一个靶向载体，包含一个由同源臂引导的nlsCre基因cDNA（nls：核定位信号），一个pgk-neo片段位于nlsCre的3'端，以及一个由MC1驱动的白喉毒素筛选基因。在RENKA胚胎干细胞中进行同源重组后，靶向克隆将cre cDNA插入Cspg4的起始密码子之后。ES细胞克隆被注入8细胞的CD1胚胎中，以产生生殖细胞嵌合体，建立敲入品系。（来源：J:171287）
