在5'端带有FRT的neo cassette，包含一个5'的loxP位点，被插入到了第五外显子上游。接着在第七外显子下游，又插入了一个loxP位点。通过Flp介导的重组，neo cassette被移除，使得从第五到第七外显子区域被标记为可剪接。通过E13.5脑组织的 Western blot 分析，证实了蛋白质表达量减少了84%（数据来源：J:170581）。
