这些小鼠是通过增强子陷阱转基因技术产生的。Calb1-icre转基因载体包含了来自小鼠Calbindin 1基因座的300bp最小启动子、经过哺乳动物密码子优化的cre重组酶（icre，无潜在剪接位点、突变的终止密码子以降低在哺乳动物中发生表观遗传沉默的可能性，以及减少 CpG 序列以减少沉默风险）、以及SV40晚期polyA信号。这个转基因被注入C57BL/6J小鼠胚胎中。最初的杂交品系与C57BL/6个体杂交。这些杂交后代又被与B6-Gt(ROSA)26Sortm1Sor小鼠杂交，以评估cre活性和遗传。之后，Cre阳性、lacZ阴性的个体与C57BL/6小鼠杂交，产生了21614品系。（来源：J:150856）
