The Ple5-lacZ transgene (bEMS90) was designed with the Ple5 MaxiPromoter (AMOTL1-X; derived from the BAC RP11-936P10 that contains the whole Angiomotin like 1 (AMOTL1) gene (along with ~60 kb of 5' and ~10 kb of 3' flanking sequences)) upstream of a frt-flanked beta-galactosidase (lacZ) gene, an SV40 early polyA signal, and a human HPRT complementary sequence (containing exon1, intron1, exon2, and part of intron2). This construct was targeted as a single copy knockin to the Hprtb-m3 mutant locus on the X chromosome. (J:144244, J:164356)
