一个由cre-loxP-neo-loxP同源序列簇修饰的向量，以及两个短同源序列簇围绕的细菌EL250，与经过修改的BAC（RP23-16G10，包含Mcpt8基因位点）一起电穿孔导入。通过同源重组，cre重组酶的编码序列被直接插入到Mcpt8起始密码子后面。通过在阿拉伯糖存在培养基中培养，删除neo cassette。然后，BAC克隆被注入C57BL/6卵母细胞的前核中。两个不同克隆源的整合BAC拷贝数通过定量PCR检测Mcpt8基因（在第1条线中有5个拷贝，第2条线中有7个拷贝）（J:164432）。
