一个反向倒置的条件性内含子(COIN)，包含lox66_SA-Egpf-polyA_lox71序列，通过同源重组以反义方式插入到exon 2，将exon 2分成了两个新exons，长度分别为120和103bp。两个lox位点呈互补方向，因此通过Cre介导的重组会不可逆地反转插入序列。此外，FRT引导的hygromycin-delta-thymidine kinase载体随后被插入在COIN内含子之后。Flp介导的重组移除了hygromycin载体。表达分析证实了纯合小鼠中全长mRNA的表达。(来源：J:163027)
