通过ET重组，含Runx2基因座的BAC被修改，插入了增强型Cre重组酶序列和一个由两个FRT座环绕的氨苄西林抗性基因，该基因位于Runx2基因的远端启动子(P1)的起始位置。随后，通过阿拉伯糖诱导的FLP重组，将BAC中的氨苄西林抗性元件去除了。线性构建被注入FVB/N雌性卵母细胞的核中。建立了三个杂交瘤系，其中一条因其表达模式被选中用于实验（J:160669）。
