这些MHCminTetO-Capn1转基因小鼠是通过将小鼠Calpain 1基因的完整编码序列插入到MHCminTetO启动子构建中产生的。构建的表达由tetO驱动，由小鼠α肌动蛋白重链(α-MHC; Myh6)启动子控制，后面跟着人类生长激素的polyA信号。该构建还包含3个修改过的GATA位点和2个改变的甲状腺样响应元件(TRE)，以维持心脏特异性的低水平表达和减少背景。总共生成了3个品系(L1、L2和L3)，表达水平逐级升高。(来源：J:125131, J:133797)
