BAC homologous recombination was used to insert cre cDNA (fused with a nuclear localization signal (NLS) sequence) immediately upstream of the initiation codon of the Ppp1r2 gene in BAC RPCI-24-322B4. The cre cDNA was followed by the bovin growth hormone-derived polyadenylation signal. In one clone, a 12-kb deletion introduced around the initial methionine codon of the apolipoprotein D (Apod) coding sequence, located 50 kb upstream of Ppp1r2, was identified. This clone was used to prevent overexpression of Apod in the brain of transgenic mice. (J:156785)
