一段人类淀粉样β（A4）前体蛋白（APP695）的cDNA序列被修改，加入了瑞典型家族阿尔茨海默病（FAD）的APP KM670/671NL突变。生成的APPswe构建随后被插入到小鼠胸腺细胞抗原1，theta（Thy1）的启动子下游（通过在小鼠Thy1基因座的exon 2的ATG起始位点插入）。这个转基因产物被注入了(C57BL/6xCBA)F1的初级卵母细胞。产生了两个创始系，其中一条显示出大脑中人类APP表达增加3倍，进行了进一步的研究。（来源：J:105265）
