The CD68-rtTA-M2 transgene was designed with a 2.9 kb promoter sequence from the human CD68 gene followed by a 83 bp fist intron (IVIS), a modified reverse tetracycline-regulated transactivator (rtTA) gene (from plasmid pUHRT-62-1), and bovine growth hormone polyA tail. This 4.5 kb transgene was microinjected into the male pronuclei of two cell (C57BL/6 x CBA) mouse zygotes. Founder line 3 (CD68-MP3) was established and backcrossed to C57BL/6 mice for at least 11 generations. The insertion site has been mapped to Chr 1. (J:154457)
