通过细菌重组酶构建的靶向载体替换了Hes1开放阅读框的大部分，其与一个由Frt内切酶切割的诺卡因抗性基因座（cre/ERT2序列）相连。在R1胚胎干细胞中进行同源重组后，产生了生殖细胞嵌合体。随后，通过与Gt(ROSA)26Sortm1(FLP1)Dym小鼠杂交，体内去除了neo cassette。(来源：J:154982, J:169830)
