在β-geo报告基因的上游，插入了一个包含FRT、F3、loxP和Lox5171序列的条件准备型基因陷阱 cassette，接着是反向排列的loxP、Lox5171、FRT和F3位点，最后在exon 1内。这个插入保持了原基因捕获的构型，导致转录在β-geo报告基因处停止，产生一个无功能的null等位基因，可通过Flp或Cre介导的去失来恢复功能。来源：(J:236978)
