在exon 4的上游插入了loxP位点和frt序列附带的neo cassette，接着在exon 4的下游也插入了另一个loxP位点。随后通过Cre酶介导的重组，去除了exon 4，留下一个loxP位点。 Western blot分析证实了纯合突变体肝脏中没有蛋白质表达。（来源：J:153089）
