A floxed neo cassette was inserted upstream of exon 8 and an additional loxP site was inserted downstream of exon 8 via homologous recombination. Cre mediated recombination then removed the neo cassette and exon 8. Removal of exon 8 results in a frame shift creating a premature stop codon at nucleotide 45 in exon 9. Western blot analysis confirmed the absence of protein expression in homozygous embryos. (J:152840)
