从Myh6基因内含子中提取的一个DNA片段，包含micro RNA编码序列，通过同源重组被一个floxed neo cassette替换。随后，通过Cre介导的重组移除了neo cassette，留下一个在内含子中的loxP位点。PCR分析证实Myh6基因的剪接模式没有受到影响。Northern blot分析证实了纯合小鼠心脏中micro RNA的表达缺失。(来源：J:152696)
