这个等位基因是由将一只雄性自然杀伤T细胞的细胞核（C57BL/6 x 129/Sv）F1背景的细胞移植到去核的卵细胞中创造出来的。随后，培养出一个囊胚并移植到伪孕鼠体内。在移植的细胞核中，有一个Tcrb基因座发生了有效的、框内重组，具体是Vbeta8S2-Dbeta1-Jbeta2S5基因片段，正是这个变异的等位基因得以延续。通过序列分析，确认这个有生产性的重组发生在C57BL/6父本遗传座上。（来源：J:152069）
