The TRE/IFN-gamma transgene was designed with the PhCMV*-1 tetracycline-responsive promoter upstream of a mouse IFN-g cDNA sequence. The PhCMV*-1 tetracycline-responsive promoter contains the human cytomegalovirus (hCMV) minimal promoter fused to the tet-operator (tetO; TRE). The IFN-g cDNA is flanked by 40 bp of 5' and 57 bp of 3' UTR sequences and includes the translation start and stop signals. The sequences that confer IFN-g mRNA instability have been eliminated from the IFN-g cDNA sequence in an effort to increase steady-state mRNA levels derived from the transgene. This 2.3 kb transgene was microinjected into fertilized B6D2F1 oocytes. Founder line 184 was established. (J:94092)
