cre/ERT2基因的cDNA被克隆到了一个2.4kb的Prrx1启动子下游。接着，一段编码IRESEGFP的cDNA被插入到了cre/ERT2序列之后。这个构建被注入到受精卵中，筛选出了3个在四肢中表达转录本的后代。与Rosa26-lacZ报告型小鼠的杂交确认了两条在用Tamoxifen诱导时有强烈X-gal染色的品系。其中一条被选中进行进一步研究。（来源：J:151681）
