Using RMCE, an inserted hygro cassette was replaced with a neo cassette and a short hairpin RNA driven by the human U6 promoter that targets Lrrk2. Expression of the shRNA results in an 82% decrease in endogenous Lrrk2 transcript expression in the brain. (J:151155)
