The gene trap vector inserted into intron 19. RT-PCR analysis indicated a more than 400-fold reduction in expression of wild-type transcript in homozygous mice. Immunoblot analysis failed to detect wild-type protein expression in MEFs from homozygous mice. The fusion protein consisting of the first 19 exons fused to the beta-geo reporter is detected in heterozygous and homozygous cells. The fusion protein is coimmunoprecipitated with SEC24A in MEF lysates. (J:186391)
