The tetracycline transactivator (tTA) coding sequence is driven by the 2.3-kb promoter from the mouse collagen, type 1, alpha 1 gene, which drives expression specifically in mature osteoblasts and osteocytes of long bones and calvariae. In the absence of tetracycline, expression of a second transgene whose promoter is regulated by the tet operator sequence (tetO) is induced; this induction is suppressed by tetracycline administration.
